a-Protease

a-Protease is an alkaline serine protease which can be used for rapid preparation of DNA from tissues, fluids, blood and cell samples. a-Protease requires Ca⁺⁺ at elevated levels for stability and can be inactivated by the addition of EDTA to chelate Ca⁺⁺ at elevated temperatures. a-Protease is stable through multiple freeze/thaw cycles. It has a functional pH range of 8.2 - 8.8. Although stable for at least six months at 4^oC, -20^oC is recommended for long term storage as a bacteriostatic measure.

Stored in 10mM TES(pH 7.0), 5mM CaCl2, 0.01% Triton X-100, and 10% glycerol.
(Unit assay conditions are 50mM HEPES-NaOH(pH 8.0), 2mM CaCl₂, 0.2%(w/v) azocasein, 1 unit enzyme, 10 minutes at 75^oC. ) 

Applications: Rapid DNA preparations for PCR, PFGE, RE analysis; inactivation of endogenous nucleases, removal of associated proteins to release DNA.
